Selective Enrichment of Hypermethylated DNA by a Multivalent Binding Platform

نویسندگان

چکیده

The preselection of hypermethylated DNA (hmDNA) from liquid biopsy samples is key to enable early-stage cancer diagnostics. Due limited selectivity the existing approaches, however, wide integration in clinic currently prohibited. Here, it argued that an affinity method on a surface, such as used chromatography, can be significantly improved by employing principles multivalency and superselectivity. In here proposed method, methyl binding domain (MBD) protein immobilized at surface receptor for (hyper)methylated DNA. By organization multiple MBDs multivalent platform achieved. MBD density increase enrichment hmDNA single binds both methylated non-methylated with small difference affinity. When varied, analyte typically responds non-linear fashion, which phenomenon called careful tuning density, envisaged over optimized. Strong applicability foreseen medical setting implementing step analytical process or lab-on-a-chip device.

DNA condensation by multivalent cations.

In the presence of multivalent cations, high molecular weight DNA undergoes a dramatic condensation to a compact, usually highly ordered toroidal structure. This review begins with an overview of DNA condensation: condensing agents, morphology, kinetics, and reversibility, and the minimum size required to form orderly condensates. It then summarizes the statistical mechanics of the collapse of ...

Thermodynamic characterization of the multivalent binding of chartreusin to DNA.

Characterization of the thermodynamics of DNA- drug interactions is a very useful part in rational drug design. Isothermal titration calorimetry (ITC), differential scanning calorimetry (DSC) and UV melting experiments have been used to analyze the multivalent (intercalation plus minor groove) binding of the antitumor antibiotic chartreusin to DNA. Using DNA UV melting studies in the presence o...

Multivalent DNA-binding properties of the HMG-1 proteins.

HMG-I proteins are DNA-binding proteins thought to affect the formation and function of transcription complexes. Each protein contains three DNA-binding motifs, known as AT-hooks, that bind in the minor groove of AT tracts in DNA. Multiple AT-hooks within a polypeptide chain should contact multiple AT tracts, but the rules governing these interactions have not been defined. In this study, we de...

Bridging lectin binding sites by multivalent carbohydrates.

Carbohydrate-protein interactions are involved in a multitude of biological recognition processes. Since individual protein-carbohydrate interactions are usually weak, multivalency is often required to achieve biologically relevant binding affinities and selectivities. Among the possible mechanisms responsible for binding enhancement by multivalency, the simultaneous attachment of a multivalent...
